Comparison of the effect of saliva on neutrophil death between healthy individual and patients with periodontal disease

Background & Aim: A conciderable percentage of neutrophils in saliva has undergone degenerative changes and the cause and mechanism of their death has not been known. Therefore the aim of this study was to determine the mechanism of the death of neutrophils which is induced by the saliva of healthy individuals and patients with peridontal diseases.

Method & Materials: For this purpose, 45 saliva samples were taken from 15 healthy individuals, 15 patients with gingivitis and 15 patients with chronic periodontitis. The saliva samples were frozen at -70°C immediately. After collecting all of the samples, the diluted saliva samples were mixed with peripheral blood neutrophils. Then staining was carried out with Annexin-V- Flous Labeling Solution and then was observed under the flourescence microscope in order to determine vital, apoptotic and necrotic cells.

Results: There were significant differences regarding the number of vital and apoptotic cells after adding saliva. There were also significant correlations between periodontal disease and the percent of vital, apoptotic and necrotic cells.

Conclusion: It is concluded that the whole saliva of patients with chronic periodontitis and gingivitis could induce neutrophil death, so the first line of defense against bacterial invasion could be broken, therefore the bacteria can remain alive and active.
